Title :生体反応を標的としたDNA切断分子の合成研究

Abstract :In recent year, it was come to light that many diseases were caused by factors of the genome. The drugs that target at abnormal protein were expected to show very effective therapeutic performance for some genomic condition occurred unusual protein expressed from exceptional genome. Inhibition of transcription factors or these relative proteins caused some genetic disease leads into development of therapeutic agents because of requiring of the right transcription from DNA to RNA in normal protein expression. We pondered that sequence specific DNA cleavage in biologically important site is practicable by synthesizing DNA cleaving molecules with the functional linker to add on DNA binding protein, for example transcription factor, in vivo. These compounds was expected to display biological activities in vivo and to use biological tools resolving some function of DNA binding proteins. So we designed the DNA cleavage compounds based on the structure of antitumor antibiotics Bleomycin to answer this purpose. The following is a summary of this study.
