Abstract : The initiation of chromosomal DNA replication must be tightly regulated, and coordinated with cell division, to replicate the genome just once per cell cycle. To understand this regulation, it is important to study the structure and function of the initiator of chromosomal DNA replication. Origin recognition complex (ORC), a sixprotein complex (Orc1p to Orc6p), is the most likely initiator of chromosomal DNA replication in eukaryotes. Although ORC of Saccharomyces cerevisiae has been studied extensively from biochemical and genetic perspectives, its structure remains unknown. Previous studies suggested that ORC has functions other than DNA replication, such as gene silencing, but the molecular mechanisms of these functions remain unknown. In this study, I used yeast two-hybrid analysis to search for ORC-binding proteins and to examine interaction between ORC subunits.
